<div><div><div><p><strong>Our immune system is incredibly powerful: among other things, it can recognize and destroy cancer cells. But tumors can be just as effective at avoiding detection – they build “invisibility shields” to protect themselves from attack.</strong></p></div></div><div><p>One breakthrough in cancer therapy has been immune checkpoint inhibitors (ICIs), drugs that reactivate immune T cells and help them fight tumors again. However, only about 20–50% of patients respond well to this treatment. Researchers are therefore seeking alternative therapeutic strategies. A small and well-known molecule called adenosine plays a key role in these efforts.</p><p>When tumor cells are stressed or dying, they release ATP, which is normally a signal that something is wrong in the tissue. But in the tumor microenvironment, ATP gets rapidly broken down by two enzymes, CD39 and CD73. Released adenosine acts like a chemical “calm-down” signal, binds to immune cells (T cells, NK cells, and others), and tells them to slow down, reducing their ability to kill cancer cells. Targeting CD73 therefore represents a promising strategy for developing new cancer treatments.</p></div><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/UOCHB_Nove_latky_mohou_podporit_imunitni_system_v_boji_s_rakovinou_88c5e82cd5.jpeg" alt="IOCB Prague: Development of potent inhibitors to boost the immune system's ability to fight cancer" srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_UOCHB_Nove_latky_mohou_podporit_imunitni_system_v_boji_s_rakovinou_88c5e82cd5.jpeg 245w," sizes="100vw" width="661" height="363"></p><p>At IOCB Prague, interdisciplinary teams led by Jan Řezáč, Jan Konvalinka, and Michal Hocek used a newly developed high-throughput screening method to test over 5,000 compounds, identifying two ribonucleoside derivatives as promising CD73 inhibitors.</p><p>The scientists then improved the compounds’ activity by carefully modifying the ribonucleoside scaffold with bulky (het)aryl groups. Further optimization improved the pharmacokinetic properties of these compounds, yielding inhibitors with low clearance, long half-life, high solubility, and strong selectivity over related enzymes such as CD39.</p><p>In cellular experiments, the compounds reduced adenosine production in cancer cells, restored T cell activation, and showed no toxicity toward human fibroblasts. We have designed and systematically studied diverse 2-substituted 7-deazapurine ribonucleoside 5′-<i>O</i>-bisphosphonates bearing a variety of (het)aryl groups at position 6 and discovered their highly potent and selective CD73 inhibition activity. The most active compounds (with single-digit picomolar <i>K</i><sub>i</sub>) contained bicyclic (het)aryl groups at position 6 in combination with chlorine at position 2. Further optimization of pharmacokinetic properties identified inhibitors with low clearance, long half-life, high solubility, and excellent selectivity over CD39 and NTPDase3. They effectively suppressed adenosine formation in MDA-MB-231 cells, rescued CD8<sup>+</sup> T cell activation, and were nontoxic to human fibroblasts. Overall, their profile compares favorably with <strong>AB680</strong>, a CD73 inhibitor currently in phase I/II clinical trials.</p></div>

Dr. Tague received his Ph.D. from the University of Utah in Physical Chemistry and his B.S. also in Chemistry from the University of Texas at San Antonio. He also conducted his postdoctoral research at the University of Virginia working with Professor Lester Andrews. He is a member of the American Chemical Society, Society for Applied Spectroscopy, American Physical Society, and the Optical Society of America. Optimization of Fluorescence Signals

This Workshop will focus on the basics of the Cary Eclipse fluorescence software and its operation as well as the optimization of the observed signal.

<p><strong>Monitoring the quality of hydraulic and/or lube oils is critical to prevent premature wear of any kind of machinery. In this session we will look at this topic from a new perspective focusing on the combination of titration (potentiometric, thermometric, coulometric Karl Fischer titration) and Vis-Near-Infrared Spectroscopy (Vis-NIRS).</strong></p>
<p>Combining titration (highly sensitive, specific, a widely used primary method) with Vis-NIRS (fast, non-destructive, results for multiple parameters at once) can make your oil condition monitoring and/or lubricant analysis much more efficient, especially, if your laboratory has to analyze large sample series every day.</p>

<p><strong>Microplastics pollution in watersheds was reported as early as the 1970’s. Now microplastics are one of the prevalent constituents of marine pollution. Microplastics are differentiated from other plastic particles, being in the size regime of &lt;5 mm. These small plastics present a potential threat to marine ecosystems due to ingestion of the microplastics but have also been noted as a means of concentrating toxic pollutants that may then enter humans through consumption of marine animals. The first step to understanding the nature of a microplastics pollution problem is the correct identification of the plastics. Fourier transform infrared (FT-IR) spectroscopy is a primary analytical method for the identification of organic materials. FT-IR microspectroscopy is well-suited to analyze particles &lt; 5 mm in dimension through the combination of the focusing power of a microscope with the chemical identification capabilities of FT-IR spectroscopy.</strong></p>
<p>We present the FT-IR microspectroscopic analyses of microplastics collected from water samples and living oceanic organisms as a means of tracing the microplastic contamination back to the originating source.</p>

Most of his career has been related to pre- and post-sales support of FTIR spectrometers, microscopes, imaging systems, and thermal analytical instruments, focusing on analyzing customer samples, demonstrations, post-sales training, troubleshooting, and R&amp;D.</p>

<p>Craig has been with Agilent for over 15 years as an ICP-MS applications scientist. He has been involved with multiple type of applications for ICP-MS, including environmental, pharmaceutical, nutraceutical, semiconductor, geologic, and clinical analyses, to name a few. Previous to Agilent he worked in an environmental lab performing analysis and supervising both the inorganic and organic sections of the laboratory. In his spare time, Craig enjoys volunteering at the local marine science center, mountain biking, hiking and relaxing at the beach. Craig obtained a bachelor of science degree in chemistry from Fort Lewis College in Durango, CO.</p>
