<p><strong>The 19th Czech - Slovak Spectroscopis Conference (19th CSSC) is a proceeding based on a long-term scientific cooperation between Ioannes Marcus Marci Spectroscopic Society and The Slovak Spectroscopic Society. The meetings organized by the former Czechoslovak spectroscopic society (1949 – 1993) in the past and the continuation in this tradition in the form national Slovak and Czech spectroscopic conferences now, since 2008, exist as a joint event.</strong></p><p>The Czech spectroscopic conference was repeated every four years, while the Slovak spectroscopic conference took place biannually. Joint scientific meetings are now held every two years, alternating between the Czech Republic and Slovakia. The order may change if this conference is combined with the European Symposium on Analytical Spectrometry (ESAS). <h4><strong>The problem&nbsp;</strong></h4><p>We live in a “global village” but we are all different and we use many different languages to communicate.&nbsp;</p><p>Even when people speak the same language, the same words may be used with different meanings. See, for example, some different uses of the word “standard” in English – a normative document, a solution of known concentration or even a type of flag.&nbsp;</p><p>Even in the same area of activity misunderstandings do occur which may result in a waste of time and money, or even worse consequences.&nbsp;</p><p>When talking about laboratory tests, lack of a full understanding of key terms and inappropriate translations leaves room for different interpretations of stated requirements. For example, there have historically been a number of different but related uses of the term ‘<strong>detection limit’</strong>. This may lead to inadequate fulfilment of those requirements, thus becoming an obstacle to the production of analytical data that is fit for its intended use. A shared interpretation of concepts related to quality in measurement between laboratories and accreditation bodies is also crucial for a fair and harmonised approach to laboratory assessment across the world.&nbsp;</p><h4><strong>A common language&nbsp;</strong></h4><p>We need a <strong>common language</strong>; clear and unambiguous. We need a common vocabulary to provide consistent definitions of concepts with their associated terms. To this end, several organisations participate in a joint effort to develop and maintain an <strong>international vocabulary of metrology</strong> (VIM) [1], which aims to achieve a unique understanding of concepts related to measurements across all sectors. The VIM is a normative reference in the standards <strong>ISO/IEC 17025, ISO 15189 and ISO/IEC 17043</strong>, and is therefore a key document for all organisations seeking accreditation.&nbsp;</p><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Eurachem_You_talk_we_understand_The_way_out_of_the_tower_of_Babel_Standards_8d947dbd5f.jpg" alt="Eurachem: You talk, we understand – The way out of the tower of Babel_A common language" srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_Eurachem_You_talk_we_understand_The_way_out_of_the_tower_of_Babel_Standards_8d947dbd5f.jpg 245w," sizes="100vw" width="1126" height="499"></p><h4><strong>Is this enough?&nbsp;</strong></h4><p>For laboratory staff some problems still stand and need to be addressed:&nbsp;</p><ul><li>Many people are confused about both concepts and terms.&nbsp;</li><li>It may be difficult to understand the “formal” VIM definitions – they are short statements intended to apply to many different measurement sectors.</li><li>Translations into local languages may add to the confusion, e.g. if different terms are used in different sectors for the same VIM concept and the translator overlooks this issue.</li><li>Some substantial changes to terminology have been made in an attempt to accommodate chemical and biological measurements. Some “old” concepts still stand but have new names, e.g. “within laboratory reproducibility” vs. “intermediate precision”.&nbsp;</li></ul><p>To address these issues Eurachem has produced a guide [2] which provides an accessible reference containing simple and in context explanations of terms most relevant to analysts, with additional examples relating those concepts to chemical and biological measurements.&nbsp;</p><h4><strong>Do you understand the meaning of these terms?&nbsp;</strong></h4><p>Are you interested in a <strong>quantity</strong> or a quality? Depending on the answer, will you use a <strong>measurement</strong> <strong>procedure</strong> or an examination? Is method <strong>verification</strong> the same or less demanding than method <strong>validation</strong>? Do you need a <strong>calibration</strong> or just performance verification? Do you have a <strong>measurement standard</strong> in your laboratory? Is <strong>metrological traceability</strong> something you worry about? Learn how single particle tracking and super-resolution microscopy unravel how actin rings influence the PM of cells.&nbsp;</p><p><strong>Presenter's Abstract</strong></p><p>The compartmentalization of the plasma membrane (PM) is a fundamental feature of cells. The diffusivity of membrane proteins is significantly lower in biological than in artificial membranes. This is likely due to actin filaments, but assays to prove a direct dependence remain elusive. We recently showed that periodic actin rings in the neuronal axon initial segment (AIS) confine membrane protein motion between them. Still, the local enrichment of ion channels offers an alternative explanation. Here we show, using computational modeling, that in contrast to actin rings, ion channels in the AIS cannot mediate confinement. Furthermore, we show, employing a combinatorial approach of single particle tracking and super-resolution microscopy, that actin rings are close to the PM and that they confine membrane proteins in several neuronal cell types. Finally, we show that actin disruption leads to loss of compartmentalization. We co-developed Deep Visual Proteomics (DVP), integrating high-resolution microscopy, AI-driven image analysis, and laser microdissection with deep proteomic profiling. Here, I will present an optimized cellenONE protocol for loss-minimized tissue processing, benchmarked across all Evosep ONE Whisper Zoom gradients and three DIA methods on the timsUltra AIP. <p><strong>Dr. Susan Bird, of Thermo Fisher Scientific, will discuss how innovative approaches in metabolomics — including SQUAD (Simultaneous Quantitation and Discovery), exposomics, and alternative fragmentation techniques — are advancing the identification and quantification of small molecules. These strategies enable greater precision and depth in small molecule research, transforming how scientists study metabolites and lipids to better understand metabolism across a wide range of scientific fields.</strong></p><p>In the second half of the presentation, Dr. Melanie Bailey from King’s College London will showcase how the SIESMIC Facility is pioneering the integration of capillary sampling with LC-MS and LA-ICP-MS to achieve sub-cellular proteomics and metals analysis. She is Director of the BBSRC SEISMIC facility for single and sub-cellular omics, an open resource for bioscientists exploit new capability in single and sub-cellular live cell omics, using capillary sampling. She is a member of the UK National Ion Beam Centre management commitee (EPSRC) and is leading the development of multimodal imaging of elemental and molecular markers, through a novel system using MeV and keV energy ion beams. Her group also have expertise in non-invasive sampling via fingerprints, sebum and saliva, and the development of tools for prognostic and diagnostic markers.</p><p>Prof Bailey is an active member of the Analytical Chemistry Trust Fund, which provides seed funding for analytical scientists. She is passionate about communicating science to the public and recently obtained a Guiness World Record for fastest marathon dressed as a cell to raise money and awareness for analytical science. She is Editor in Chief of the RSC's longest running journal, The Analyst.</p><p><strong>Presenter: </strong>Susan Bird, PhD (Metabolomics Vertical Marketing Manager, Thermo Fisher Scientific)</p><p>As Sr. Manager, Susan is responsible for leading the Metabolomics vertical marketing strategy and leads both the scientific applications and vertical marketing programs. This includes initiatives to expand and strengthen strategic relationships and guide product development to meet the emerging needs of our Metabolomics customers.</p><p>Prior to this role, Susan worked in commercial as an instrument sales rep in the Boston and Cambridge area. Prior to joining Thermo Fisher, she trained as a post-doc and Instructor of medicine at Brigham and Women’s Hospital and Harvard Medical School where she developed non-targeted and targeted lipid profiling methods, applied to epidemiology cohorts.</p><p>This work used primarily LC-MS with Orbitrap detection and provided key contributions to the emerging field of lipidomics.</p></blockquote>
