<p><strong>Introduction:</strong> Introduced in Agilent ICP-MS MassHunter version 4.6, IntelliQuant for ICP-MS provides an automated semiquantitative analysis for every sample. Calibration and background correction are automatically made with no user intervention,  and data is presented in an easily understood periodic table heat map format and a comprehensive data table.</p>
<p>IntelliQuant is possible because a QuickScan can be performed when measuring each sample. The optional two second QuickScan acquires a complete mass spectrum of the entire measurable mass range, facilitating semiquantitative and Total Matrix Solids (TMS) calculations of every sample.</p>
<p>TMS levels are calculated from the IntelliQuant data. Similar to Total Dissolved Solids (TDS), TMS is the sum of the key matrix elements in each sample. Included in the TMS calculation are the elements typically considered to contribute to matrix effects, such as ionization suppression and matrix deposition on interface cones and lenses. Gas elements, including elements in acids such as HCl and HNO₃ , do not cause appreciable matrix effects in ICP-MS, so they are excluded from the calculation.</p>
<h4><strong>2. <p><strong>Introduction:</strong> Synthetic oligonucleotides are short chains (oligomers) of deoxyribonucleic acid (DNA) or ribonucleic acid (RNA) molecules. They are used in applications such as polymerase chain reaction (PCR) tests, DNA sequencing and novel therapeutic products.</p>
<p>The synthesis of oligonucleotides involves a four-step cyclic process followed by multi-step downstream purification. This manufacturing process as well as environment, packaging, and container closure systems can be potential sources of contamination leading to inorganic impurities formation.</p>
<p>To control and monitor product safety, permitted daily exposure (PDE) limits of 24 elements are defined in ICH guideline Q3D(R2)1 and USP chapter &lt;232&gt;2 . ICH Q2(R1)3 and USP&lt;233&gt; 4 describe analytical measurement requirements and procedures for the measurement of those elemental impurities. Although oligonucleotide products have not been included in those regulatory guidelines yet, the industry common practice is to follow those guidelines.</p>
<p>In this study, an analytical workflow (Figure1) was developed in accordance with the ICH/USP guidelines for the analysis of elemental impurities in an oligonucleotide sample using an Agilent 7850 ICP-MS. Data was acquired to evaluate the accuracy, specificity, reproducibility, and ruggedness of the method.</p>
<p><strong>Conclusions:</strong> The study has demonstrated the suitability of the Agilent 7850 ICP-MS for the identification and quantification of 24 elements in a synthetic oligonucleotide sample in compliance with existing USP and ICH guidelines. Instrument and method performance requirements for stability, detectability, precision, specificity, ruggedness, accuracy, and spike recovery were all achieved per the criteria given in USP &lt;232&gt;/&lt;233&gt; and ICH Q3D(R2)/Q2(R1).</p>
<p>As the development and use of oligonucleotides in therapeutic applications continue to advance, the elemental impurities analysis of oligonucleotides using 7850 ICP-MS can safeguard the product quality and safety.</p>

<p>Standard Raman microscopes are traditionally used to perform Raman measurements or Raman spectroelectrochemical experiments when they are coupled with electrochemical equipment. Raman spectra collected with confocal microscopes allows the characterization of very small areas. However, these instruments can exhibit several limitations. SPELEC RAMAN, a new generation of spectroscopic and spectroelectrochemical instruments, offers powerful and interesting features to overcome these issues. In this Application Note, a detailed comparison is made between the main features of a standard Raman instrument and the SPELEC RAMAN by analyzing the results obtained with both instruments.</p>
<p><strong>CONCLUSION</strong></p>
<p>SPELEC RAMAN exhibits a number of competitive advantages over standard Raman microscopes. For instance, the spot size of SPELEC RAMAN allows the characterization of a large area in a single experiment, obtaining representative results with only one measurement. The versatility of the Raman probe is demonstrated since it can be used with different cells. The portability as well as the simplicity of SPELEC RAMAN facilitate the performance of optical experiments. It also ensures the synchronization of electrochemical and optical signals in case Raman spectroelectrochemical measurements are carried out. In addition, DropView SPELEC software allows realtime acquisition of the data and the performance of operando measurements. The easy data treatment and analysis of the results is done using one-click tools.</p>

<p>Bioimaging and metallomics are complementary fields of study that explore the role of analytes and metals in biological systems. Bioimaging visualizes the presence or absence of certain tissues, cells, and low molecular weight compounds while metallomics seeks to understand the functions and roles of metals in biological structures. Analytical techniques such as liquid-chromatography-inductively coupled plasma-mass spectrometry (LC-ICP-MS), single cell-ICP-MS, and laser ablation (LA) ICP-MS have been used for protein quantification1 , single cell analysis2 , and bioimaging, respectively.</p>
<p>LA-ICP-MS can provide detailed information about the localization of elements on the surface of solid samples, as demonstrated in this study for the investigation of the distribution of metals on thin sections of biological samples. </p>

<p><strong>Goal:</strong> To demonstrate the capabilities of the Thermo Scientific™ iCAP™ PRO XP ICP-OES Duo instrument, in conjunction with the Thermo Scientific™ iSC-65 Autosampler, for the fast and robust elemental analysis of fertilizers.</p>
<p><strong>Conclusions:</strong> This application note presents a fast and optimized method for the quick, accurate, and robust analysis of major and trace elements in fertilizers using the iCAP PRO XP ICP-OES Duo instrument and the iSC-65 Autosampler. The highlights of the study are listed below:</p>
<ul>
<li>A quick and simple method with a run time of 36 s per sample for all target elements in fertilizers has been developed. This has been achieved using the excellent sensitivity in radial mode and the fast data acquisition capabilities of the iCAP PRO XP ICP-OES Duo instrument using the iFR mode. Due to the fully simultaneous acquisition across the full optical spectrum, an additional exposure can be eliminated, offering a 50% reduction of the time required for data acquisition. </li>
<li>The use of the Step Ahead feature available on the iSC-65 Autosampler compliments the speed of the iCAP PRO XP ICP-OES Duo instrument and the chosen settings in this method by enabling rinsing parallel to data acquisition. The combination of both provides the opportunity to reduce sample turnover times further by almost 30% compared to a standard setup.</li>
<li>The method allows achievement of the typically required limits of quantification for commonly regulated elemental contaminants in fertilizers—such as As, Cd, Cr, Hg, Ni, and Pb—with respect to the criteria prescribed in the Regulation (EU) 2019/1009. At the same time, it is capable of covering a large linear dynamic range of 0.01–20 mg·kg-1 for trace elements and 1–1,500 mg·kg-1 for elements present in higher concentrations. If the total concentration of Cr is found to be higher than the specified limit, then a speciation technique may be used to determine the amount of Cr (VI) present in the samples. The concept of the application can be easily extended to other sample types, such as soil or wastewater analysis. ICP-MS instruments can also benefit from single mode measurement and the high-speed capabilities of the iSC-65 Autosampler.² </li>
<li>Continuous, uninterrupted measurements of &gt;5 hours covering more than 500 unknown samples proves the accuracy and unmatched robustness of the developed method, which makes the iCAP PRO XP ICP-OES Duo instrument an ideal choice for high productivity analytical testing laboratories.</li>

<p><strong>Goal:</strong> To demonstrate the analysis of process water from hydrogen fuel cells for inorganic trace contaminants with high sensitivity and accuracy using triple quadrupole ICP-MS.</p>

<p><strong>Conclusions:</strong> The iCAP TQe ICP-MS was employed to analyze 53 elements including fluorine in process water derived from hydrogen fuel cells. Among the analytes, several critical interferences can cause unexpected bias, and these were investigated closely for effective and complete removal by means of selective collision / reaction cell reactions with oxygen. This study also included the development of a method for the analysis of fluorine, generally only possible in an indirect measurement. This analytical method was rigorously tested, and the results obtained clearly demonstrate the fowling analytical advantages:</p>
<ul>
<li>The iCAP TQe ICP-MS operated in TQ-O2 and He KED modes allowed the analysis of 53 elements in relevant sample matrices over a wide concentration range. For minor elements, the method was tested over 4 orders of magnitude (from 0.01 to 100 µg·L-1), whereas major elements were tested between 1 to 100 µg·L-1.</li>
<li>Using a high throughput analysis approach, with a new autosampler equipped with a Step Ahead feature for minimizing total analysis time, all elements relevant for the development of hydrogen fuel cell technology could be measured accurately with low detection limits.</li>
<li>The use of TQ-O₂ mode provided advanced performance for eliminating the polyatomic interferences of phosphorus, sulfur, silver, and cadmium in the simulated hydrogen fuel cell process water. This covered both polyatomic interferences as well as overlaps created by intense signals from isotopes of elements present in high concentrations.</li>
<li>The iCAP TQe ICP-MS offers the flexibility to choose high resolution for the Q1 and Q3 quadrupoles, enabling complete elimination of polyatomic interference and isobaric interferences. </li>
<li>The feasibility of fluorine analysis was demonstrated using an indirect method after complexation with barium. The iCAP TQe ICP-MS showed accurate and precise determination of fluorine as proven by the spike recovery tests performed.</li>
<li>Excellent stability performance was obtained with the iCAP TQe ICP-MS together with excellent CCV and spike recovery results over a 3-hour run without internal standard correction. This provides the advantage of avoiding potential contamination from addition of the internal standard solution.</li>

<p>Non-metallic inclusions dramatically affect steel properties and cause process issues like nozzle clogging. The costs associated with bad steel properties, process issues or customer complaints may amount to several hundred thousand dollars every year. Controlling the formation and the modification of non-metallic inclusions has therefore become essential to the production of modern steels as demanded by today’s markets.</p>
<p>The Thermo Scientific ARL iSpark Plus is a high-end metal analyzer that allows precise and accurate elemental analysis of steel. It also enables ultra-fast analysis of non-metallic inclusions, enabling a steel plant to control steel quality and production process even more effectively, and considerably reduce the costs associated with unwanted inclusions.</p>
<p>The Basic Inclusion Analysis Spark-DAT (Spark Data Analysis and Treatment) option is the entry-level method for standard inclusion analysis in low alloy steel. It reports the amounts of some of the most common types of non-metallic inclusions together with the elemental concentrations, helping steel plants to guarantee the quality and the properties of the steel product, as well as the smooth running of the production process.</p>

<p><strong>Abstract:</strong> The Agilent Resolve handheld Raman system is a versatile analyzer that can be deployed on location to identify a broad range of materials. It uses traditional surface analysis mode and through-barrier mode through Agilent&#39;s proprietary spatially offset Raman spectroscopy (SORS) technology. SORS enables samples that are concealed behind barriers such as colored and opaque plastics, paper, and highly pigmented paints and varnishes to be identified. This application note details how the Resolve Raman system can be used to detect concealed endangered species products using SORS. Data are presented that show the effectiveness of Resolve operating in traditional surface mode to distinguish ivory from a plastic substitute. Further analysis of endangered animal products using surface mode enabled the division of animal products into two broad categories: mineral-based and protein-based. Principal components analysis (PCA) was used to further differentiate the protein-based samples, which included pangolin scale, rhino horn, dried seahorse, and totoaba fish bladder, based on their spectral differences.</p>
<p><strong>Conclusion:</strong>  The Agilent Resolve handheld Raman analyzer includes various spectral libraries that enable the identification of a broad range of chemicals and materials. This study has shown the application of the instrument and the SORS technique for the detection of endangered animal species. Using a combination of conventional surface and through-barrier modes of analysis, Raman spectroscopy and SORS technology can be used to:</p>
<ul>
<li>Identify ivory through layers of paint and varnish.</li>
<li>Differentiate plastic substitute materials from authentic ivory and bone.</li>
<li>Distinguish between mineral-based animal products, such as ivory and armadillo scale, from protein-based animal products, such as rhino horn and pangolin scales.</li>
</ul>
<p>The simple-to-use Resolve handheld Raman analyzer with SORS capability can provide complimentary data to help detect and identify endangered animal-derived samples.</p>

The widespread demand for glucagon-like peptide receptor agonists (GLP-1 RAs) has put pressure on synthetic peptide manufacturing capacity and revealed the need for efficient and scalable tools. The Agilent Vaya Raman raw material identity verification system enables uniquely efficient raw material verification due to its through-container identification capabilities. This application note showcases Vaya&#39;s successful through-container identification and differentiation of key building blocks for synthetic peptide manufacturing: fluorenylmethoxycarbonyl (Fmoc)-protected amino acids.</p>
<p><strong>Conclusion:</strong> The increasing demand for peptide biologics, such as GLP-1 receptor agonists, is expected to continue growing and outpacing current manufacturing capacity. To effectively meet demand, scalable solutions like the Agilent Vaya Raman raw material identity verification system introduce efficiencies to avoid bottlenecks that could be faced in raw material identification and regulatory-enforced verification. Additionally, the Vaya&#39;s ability to perform nondestructive, noninvasive identification ensures that raw materials are transferred expediently to production while preserving their integrity. </p>
