Discoveries from IOCB Prague now have a greater chance of being put into practice starting today, and patients with previously untreatable diseases have stronger hopes of receiving effective therapy. A unique translational center connecting science with its commercial applications – Pharmtheon – is now operational, marking a significant development in Czech science.
State-of-the-art laboratories have found their home at Harfa in Prague, where preclinical research will be conducted to bring promising molecules closer to the development of important medications.
&quot;If the discoveries of modern science are to serve society with practical results, centers like Pharmtheon are essential. In Prague, where we have access to a range of excellent scientific results at the intersection of chemistry and biology, such an institution has been regrettably missing. I am very pleased that the idea of Pharmtheon has been brought to fruition. It is actually our duty to build upon the previous successes of the institute in the field of medicinal applications,&quot; commented Jan Konvalinka, Director of IOCB Prague.

The transfer of technologies into practice has long been supported by the Czech government, which has identified this area as one of its priorities in its program statement. The first project to be developed at the center is the development of substances that reduce the frequency of mutations in cancer cells. These mutations are the cause of resistance to some drugs used in the chemotherapy of cancer patients. This area is highly relevant and is currently somewhat overlooked.
&quot;The development of new drugs is particularly challenging in the environment of academic institutions, where basic research is the priority. This may not align with efforts to promote the transfer of inventions into practice. Therefore, I consider the creation of a suitable environment that combines expert knowledge, financial resources, and technical equipment to be crucial. That is also the main reason why Pharmtheon was established,&quot; explained Martin Fusek, the initiator of the center&#39;s establishment.

The creation of new drugs first requires an understanding of the mechanism of disease development and then attempts to influence these mechanisms with administered drugs. If substances that can positively influence certain pathological processes at the cellular level are found, these substances need to be optimized in terms of effectiveness, side effects, duration of action, and many other properties in preclinical research. This process is called translational research. Now, thanks to Pharmtheon offering professional support to translational research and enabling more efficient collaboration among all involved parties, even better results can be expected.
Pharmtheon Center is part of the IOCB Tech family, a subsidiary of the Institute of Organic Chemistry and Biochemistry of the Czech Academy of Sciences in Prague.

From individual cells and small tissue samples to fruit flies: if you want to research biological material, you often need very special equipment. The <a href="https://www.tuwien.at/en/koop/amt/projects/lifescope-3d">&quot;LifeScope3D&quot; platform</a> has now been set up at TU Wien to provide optimal instrumentation for studying 3D multicellular structures, such as organoids and spheroids. A whole range of specialized equipment is now available at the Getreidemarkt campus, which enables characterization on various scales- starting from entire objects down to single cell or molecule analysis. However, the platform is not only interesting for biological samples, but also for other similar-sized structures, such as those produced by a high-resolution 3D printer.
Molecular biologist, Mina Petrovic, PhD, is the project manager of the new platform. LifeScope3D is the result of a collaboration between Prof. Aleksandr Ovsianikov (Institute of Materials Science and Technology), Prof. Philipp Thurner (Institute of Lightweight Structures and Structural Biomechanics) and Prof. Gerhard Schütz (Institute of Applied Physics). However, the platform will also be accessible to all other research groups - both within and outside TU Wien. LifeScope3D is intended to create a hub that promotes multidisciplinary collaborations in the field of cellular assemblies and tissue engineering. This was made possible by funding from the Austrian Research Promotion Agency FFG.

<h4>Different measuring devices</h4>
&quot;We can not only image biological structures, but also fully analyze them in three dimensions,&quot; explains Mina Petrovic. &quot;This is of great importance for many applications in biology.&quot; For example, a special microscope on the new research platform can be used to illuminate the samples with so-called &quot;lattice light sheets&quot;. Instead of a beam of light that scans the sample point by point, an extremely thin, two-dimensional light sheet is generated that penetrates the sample layer by layer. Each time, only dyes located within this thin light sheet are detected and a three-dimensional image with high contrast can then be put together on the computer.
However, optical methods are not the only way to examine samples - mechanical experiments also play an important role. The sample can be subjected to a precisely defined mechanical load. While the sample is compressed, the sample deformation is measured. Valuable findings can be derived from this, as such experiments mimic the pressures that cells have to endure in different parts of the body.
A bio-sorter is also used in the new research platform: &quot;Biological samples are rarely uniform, but often come in a mixture of different sizes, showing different features”, explains Mina Petrovic. &quot;In the bio-sorter, a mixture of, for example, multicellular spheroid structures is guided past a laser beam where each individual structure can be precisely analyzed and categorized at a very high speed.&quot;

<h4>Accessible for everybody</h4>
Numerous research groups from different faculties at TU Wien are conducting research on biological samples - LifeScope3D will open up new analysis methods for all of them. However, the aim is also to collaborate with research teams outside of TU Wien: &quot;In principle, LifeScope3D is open to all groups with suitable research projects,&quot; says Mina Petrovic. &quot;Anyone who wants to use our equipment can get in touch with us and arrange suitable timeslots; we will then also provide advice and help with the correct execution of the experiments.&quot;
The research is not necessarily limited to biological materials. &quot;The devices are ideally suited for biological analyses, but who knows - we may also receive exciting research requests from other disciplines that we might not even be thinking about today,&quot; says Mina Petrovic.

TU Wien launches a research platform for 3D testing of bio-samples

Environmental analytical laboratories play a key role in protecting the environment, and it is therefore essential that they themselves take action towards sustainability. This EnviroMail focuses on the annual plastic savings that one unit of European laboratories have achieved through innovative approaches and changes in working practices. Specific measures such as replacing single-use plastics with reusable alternatives, optimizing processes and using modern technologies were used for plastic saving. Our achievements show that SUSTAINABILITY is not just a word for us, but we are continuously working on a greener future.&nbsp;In recent decades, plastic has become an integral part of our lives. Its practical properties such as low weight, durability, and low cost have led to its global mass use. However, the increase in plastic waste, especially single-use plastics, has caused significant environmental problems. In the laboratory, we also use a lot of laboratory utensils and equipment made of plastic. Most of them end up as hazardous waste after the use. Reducing the amount of plastic consumables used has become one of our company's key sustainability issues.<h3>Problems with plastics</h3>Plastics, particularly single-use items like plastic bottles, bags, packaging, and straws, are a major source of waste that pollutes our oceans, rivers, and land. This waste is highly resistant to decomposition, leading plastic items to persist in the environment for decades or even hundreds of years. Microplastics, tiny pieces of plastic, are not only found in the natural world but also in the food chain, posing a risk to both human and animal health.<h3>Sustainable approaches to plastics saving</h3>In the fight against plastic waste, there are a number of initiatives aimed at saving plastic and promoting sustainable development. Key approaches include:<ol><li>Reducing plastic consumptionReducing the consumption of plastics, especially single-use plastics, is a key part of the solution to the plastic problem. This includes reducing the use of plastic consumables, or at least using smaller sizes of different containers and tubes that are then lighter in weight or substituting other materials.</li><li>Plastic recyclingPlastic recycling is another key tool in the fight against the plastic waste. Improving recycling processes, introducing new technologies and promoting recycled materials in industry can help to significantly reduce plastic waste. In addition, recycling plastics saves natural resources, reduces CO2 emissions and helps to save energy.</li><li>Alternatives to plasticsThe development of alternative materials to plastics is a further step towards sustainability. For example, bioplastics, made from renewable raw materials such as corn or potatoes, are a greener alternative to traditional plastics. Some companies are also focusing on creating packaging that is compostable or easily recyclable.</li></ol><h3>Towards ALS Net Zero 2050</h3>Saving plastics is a key to achieving the Sustainable Development Goals. In one of the main European laboratories, it was managed to reduce plastic waste production by 1,758 kg in the last financial year 2023/2024. When the weight of plastic is taken into consideration, the saving in amount of waste is significant.<h3>Plastics saving areas:</h3><ul><li>Use of smaller disposable plastic test tubes. Total savings: 915 kg</li><li>Replacement of plastic containers with containers made of bioplastic. Total savings: 412 kg</li><li>Ensuring recycling of used plastic products. Total savings: 152 kg</li><li>Replacing plastic packaging with paper packaging that is more easily recycled. Total savings: 145 kg</li><li>Reducing the consumption of plastic containers. Total savings: 58 kg</li><li>Purchase of non-packaged laboratory syringes for water sample filtration. Total savings: 47 kg</li><li>Discontinuation of the purchase of bottled water. Total savings: 20 kg</li><li>Implementation of electronic documentation management system, reduction of EURO packaging consumption. Total savings: 10 kg</li></ul><h3>Conclusions</h3>Saving plastics is an essential step towards protecting the environment and promoting sustainable development. It is a challenge that requires a coordinated effort by all. Reducing plastic consumption, improving recycling processes, promoting alternative materials and changing consumption habits are key factors in ensuring a sustainable future for our planet. By continuing to work towards these goals, we can help to protect natural resources, improve health and ensure a good quality of life for future generations.

EnviroMail™24/Europe: Saving Plastics for Sustainable Development in ALS Laboratories

Proper sampling process, along with the selection and utilisation of appropriate sampling containers, represent two critical steps prior to the commencement of laboratory analyses that significantly influence the accuracy of environmental analyses and, consequently, the final reported results. Such system incorporates regular measurement of quality control samples such as blank samples, duplicates, and laboratory control samples. However, less attention is often paid to ensuring quality during initial sampling and transport of samples to the laboratory. These processes are integral to the analytical procedure, with their impact occurring before the samples reach the laboratory. Inappropriately collected or contaminated samples can lead to inaccurate or misleading data and information. The general requirements for water sampling are specified in the international standard <a target="_blank" rel="noopener noreferrer" href="https://www.iso.org/standard/82273.html">EN ISO 5667-3: 2024 "Water quality - Sampling - Part 3: Preservation and handling of water samples"</a>. This document outlines and describes basic definitions and terms, as well as requirements for sampling containers, transport, identification, and storage of samples.&nbsp;<h3>ALS Sampling Containers</h3>ALS laboratories continuously work to improve laboratory service. ALS laboratories routinely follow scientific publications, national and international standards and perform testing of sampling containers, including stability studies. All sampling container batches provided by ALS are regularly tested to detect any unexpected changes during their manufacturing process or distribution. All standard sampling containers provided by ALS are intended for single-use only to ensure the quality of provided containers.<h3>Selection of sampling containers</h3>The selection of an appropriate container can have a significant impact on the measured results. Typical phenomena include sorption of the analyte, contamination of the sample, or possible loss of the analyte of interest. Therefore, it is essential to ensure and verify the suitability for the intended purpose:&nbsp;<ul><li>Suitability of the material (plastic/glass)&nbsp;</li><li>Chemical resistance</li><li>No adsorption for specified analyte</li><li>Purity of the material</li><li>Leak-proof / airtight container</li></ul>An important parameter is also the volume/size of the sampling container. ALS laboratories emphasize the miniaturisation of sampling containers. If a sampling container contains a chemical preservative, it should never be rinsed prior to use. Chemical preservation is an effective tool that significantly extends the stability of target analyte and thereby the quality of results. The chart shows stability study for the determination of ammonium ion using an identical sample (effluent from a wastewater treatment plant; WWTP) with and without a chemical preservation. Sample without preservation must be processed within 48 hours, otherwise there is a significant decrease of target parameter, thus false negative results. ALS Laboratories are one of the few in the world to offer commercial isotope ratio testing services not only for radiogenic systems (Sr, Nd, Pu, U) and light stable isotopes (Li, B, Si), but also for heavy stable elements such as Ag, Ca, Cd, Cu, Fe, Mg, Mo, Si, and Zn. Most people are familiar with radiocarbon dating, perhaps the most famous use of isotope ratio analysis, where the ratio of radioactive Carbon-14 to Carbon-12 isotopes can accurately determine the age of organic materials up to around 60,000 years. Isotope Ratio Mass Spectrometry (IRMS) is widely used to measure stable isotope ratios of the most common light elements, including C, N, S, O, and H. Measurement of stable isotope ratios of heavier elements tends to be more challenging, requiring specialized instrumentation, and typically requiring preconcentration, because heavy element concentrations are very low in most sample types. Our expert team has made substantial contributions to the field of isotope ratio analysis with 170 peer-reviewed publications. ALS laboratories offer isotope ratio analyses for more than 20 stable and radiogenic isotopic systems in a variety of sample matrices with high precision, even where sample concentrations are very low.Most isotope ratio tests require high precision, because the differences observed in isotope ratios for most elements are small. MC-ICP-MS is used for the most demanding isotope ratio measurements, where the best possible precision is required. For example, age dating of rocks and meteorites using isotopic ratios of Samarium and Neodymium requires a precision of at least 0.002%, which can only be achieved with MC-ICP-MS. The uncertainties with ICP-SFMS are higher, ranging from about 0.05-1% (depending on test), but are sufficient for some applications. Test results for isotopic ratios are typically reported as delta (δ) values in units of parts per thousand (‰), which relate to internationally accepted reference standards for each isotope system.<h3>Isotope Ratio Testing Requirements and Options</h3>Isotope ratio tests are available for a wide variety of matrices, including (but not limited to) natural and process waters, wastewaters, soils, sediments, aerosols, vegetation, biota, food products, clinical samples, archaeological objects, metals, and alloys.Isotope ratio tests and application requirements are complex. All interested experts from ALS can discuss requirements with our clients to determine the most appropriate options, which may require customized sample preparation techniques such as matrix removal, analyte preconcentration, and purification in addition to IRMS analysis.
